Use of Polymerase Chain Reaction To Detect the Take-All Fungus, Gaeumannomyces graminis, in Infected Wheat Plants

摘要: Gaeumannomyces graminis, the causative agent of take-all disease wheat, barley, and oats, was detected in infected wheat seedlings by using polymerase chain reaction to amplify Gaeumannomyces-specific DNA fragments. Nested primers two rounds amplification were used fragments, approximately 287 188 bp size, from G. graminis-infected seedlings. The use nested greatly decreased number nonspecific products. Polymerase products not obtained with several other phytopathogenic fungi or uninfected Amplified visualized on agarose gels, their identities confirmed hybridization. This method did require culturing fungus has potential for detecting graminis infested oat fields.
