摘要: A human telomere DNA segment (HT-DNA) can fold into a G-quadruplex in the presence of some monovalent cations. These cations interact with phosphate groups and/or O6 oxygen atom guanines, which are called non-specific interactions and specific interactions, respectively. However, until now how these two affect structure HT-DNA has not been well understood. In this study, combination analytical ultracentrifugation (AUC) circular dichroism (CD) was used to explore effects on 22-mer single-stranded sequence 5′-AGGG(TTAGGG)3-3′. The results showed that standard sedimentation coefficient (s20,w) starts increase when concentration potassium ions (CK+) is higher than 10.0 µM due formation through interactions. Whereas, for control DNA, CK+ value 1.0 mM needed increasing s20,w Moreover, could promote much more easily lithium, sodium cesium ions, presumably its appropriate size dehydrated state easier dehydration. molar mass at different cation concentrations nearly constant close theoretical monomeric HT-DNA, indicating what we observed structural change individual chains.
