摘要: Publisher Summary This chapter focuses on the analysis of circular dichroism (CD) data to determine thermodynamic parameters folding, binding constants, and estimates secondary structure. Proteins polypeptides have CD bands in far ultraviolet region that arise mainly from amides protein backbone are sensitive their conformations. near visible regions, which aromatic amino acids prosthetic groups. can be used enthalpy, entropy midpoints, values unfolding/refolding transitions a if they reversible as function temperature or denaturant. The change ligand concentration has been study numerous systems. It is found two proteins bind each other only when folded complex unfolds cooperatively reversibly give unfolded monomers, it easy constant by determining thermodynamics folding compared with monomers.
