摘要: Fast-scan cyclic voltammetry has been used to measure dopamine (DA) synaptic overflow in slices of rat caudate nucleus induced by electrical stimulation with one-, two-, and 50-pulse, 10-Hz trains. Synaptic this preparation is shown be the result competing effects release cellular uptake. Release caused all pulses was attenuated D2 agonist quinpirole (1 microM). The rapid time response measurements (100 ms) allows autoinhibition endogenous, released DA resolved real time. concentration during second pulse a train 58% that first pulse, an effect partially blocked addition 2 microM sulpiride, antagonist, perfusion buffer. stimulus unaffected suggesting autoreceptors are not normally occupied preparation. third 14% also could enhanced sulpiride. duration inhibition endogenous estimated varying interval between one-pulse stimulations until equal on first; half-time approximately 17 s found. picrotoxin microM) glutamate (10 buffer did affect stimulated DA, although atropine for trains tested.
