Importance of the rice TCD9 encoding α subunit of chaperonin protein 60 (Cpn60α) for the chloroplast development during the early leaf stage

摘要: The chloroplast development governs plant growth and metabolism. This study characterized a novel rice thermo-sensitive 9 (tcd9) mutant, which exhibited the albino phenotype before 3-leaf stage grown below 24 °C whereas displayed normal green at over 28 or even 20 after 5-leaf stage. obvious decrease in Chl levels, abnormal chloroplasts with few thylakoid lamella thylakoids were observed for mutant seedlings low temperature, but was apparently high suggesting thermo-sensitivity of phenotype. Genetic analysis showed that controlled by single recessive nuclear gene (tcd9). map-based cloning molecular complementation tests revealed mutation TCD9 encoding α subunit Cpn60 protein (Cpn60α), localized chloroplasts, responsible In addition, expression all tested tissues, especially young leaves. transcriptional indicated levels studied genes related to tcd9 seriously affected °C, some them recovered into green-seedlings 32 °C. Our observations suggest nuclear-encoded Cpn60α plays crucial role early leaf rice.
