摘要: Aphids are virus-spreading insect pests affecting crops worldwide and their fast population build-up insecticide resistance make them problematic to control. Here, we aim understand the molecular basis of spotted alfalfa aphid (SAA) or Therioaphis trifolii f. maculata in Medicago truncatula, a model organism for legume species. We compared susceptible resistant near isogenic lines upon SAA feeding via transcriptome sequencing. Expression genes involved defense stress responses, protein kinase activity DNA binding were enriched line. Potentially underlying some these changes gene expression was finding that members MYB, NAC, AP2 domain ERF transcription factor families differentially expressed versus lines. A TILLING created cultivar screened using exome capture sequencing served as reverse genetics tool functionally characterise response. This screening revealed three factors (a ERF) important regulators defence response, premature stop-codon background led delay mortality enhanced plant susceptibility. combined functional genomics approach will facilitate future development pest by uncovering candidate target can convey resistance.
