P21 ras Transforming Protein: Significance of the Carboxy Terminus

摘要: The p21 ras genes form a multigene family that is conserved in evolution (reviewed ref. 1). These were first detected and studied two rodent-derived acute transforming retroviruses, Harvey (Ha) Kirsten (KO murine sarcoma viruses (MuSV); both are highly oncogenic vivo induce tumorigenic transformation of established cell lines vitro. As true for other retro-viruses, the capacity each virus resides its viral oncogene (v-onc). v-onc Ha-MuSV Ki-MuSV v-ras gene was transduced from cellular (c) gene. 21 kd encoded proteins share antigenic determinants. However, under stringent hybridization conditions, sequences base pair with different restriction endonuclease fragments genomic DNA given animal species, indicating c-ras gave rise to genes. Molecular cloning sequencing have directly confirmed this conclusion (2–9). mammalian genome contains at least three biologically active genes: H (similar ras), K N (first human neuroblastoma line).
