摘要: Systemic lupus erythematosus, Sjogren’s syndrome, and dermatomyositis are systemic autoimmune diseases that develop after environmental triggering of genetically susceptible individuals. The precise cellular molecular mechanisms leading to disease, what factors determine which organs involved, remain poorly understood. Recent insights into genetic susceptibility now make obvious triggers often act via pathways containing disease-associated polymorphisms. In the breaking tolerance, initiating tissue— including dendritic cells—provides a decisive microenvironment aff ects immune-cell diff erentiation, activation adaptive immunity. Type 1 interferon produced by innate immune cells has central role in autoimmunity activates B T cells. turn, B-cell-derived autoantibodies stimulate produce type interferon; thus, positive feedforward loop is formed includes both systems. New treatments could simultaneously specifi cally target several such vital autoimmunity.
