摘要: B-1 cells (CD5 B cells) represent an initial fetal wave of cell lymphopoiesis. have fundamental properties that are unique from conventional cells, including a restricted Ab repertoire. We investigated the mechanism for overrepresentation one such Ig H chain variable-region gene, VH11, by murine cells. postulated cis-regulatory element contributed to use VH11. observed DNA encoding leader peptide VH11 was atypically A/T rich and thus candidate nuclear protein binding. By electrophoretic mobility shift analysis, we found specifically bound three complexes present in nucleus line AJ9. Of these bands, ubiquitous all examined (lymphoid nonlymphoid); another band only third specific expressed or VH12. In addition its binding properties, sequence also displayed modest tissue-specific enhancer activity. footprint were bind octamer motif embedded within DNA. To identify octamer-binding proteins, panel octamer-specific Abs used. Oct-1, cell-specific Oct-2. The neither Oct-1 nor Oct-2, but may be novel POU domain protein. hypothesize site target this gene preferential rearrangement and/or expression therefore would contributing factor increased
