摘要: A comparison of histochemical detection GM1 ganglioside in cryostat sections using cholera toxin B-subunit after fixation with 4% formaldehyde and dry acetone gave tissuedependent results. In the liver no pre-treatment showed detectable differences related to reaction products, while studies brain superiority preextraction (followed by formaldehyde), which yielded sharper images compared diffuse, blurred staining pattern associated formaldehyde. Therefore, aim our study was define optimal conditions for B-subunit. Ganglioside extractability acetone, ever neglected topic, tested comparing anhydrous containing admixture water. TLC analysis extractable from did not exceed 2% total content at -20°C, room temperature. The loss increased 30.5% 9:1 acetone/water. Similarly, photometric lipid sialic acid, extracted dried homogenates gangliosides into 3.0±0.3% only. homogenate 9.5±1.1%.
