摘要: Progestin hormones are vital for inducing oocyte maturation in fish by binding to membrane progestin receptors (mPRs). The aim of this study was examine the expression and regulation mPRalpha mPRbeta zebrafish follicles. First, defolliculated fully grown oocytes were subjected immunofluorescent staining using anti-mPRalpha antibodies, their on confirmed. Second, total protein collected from follicles Western blotting revealed that level increased with follicle development. We have previously shown several members transforming growth factor-beta (TGF-beta) superfamily, including TGF-beta1, activin-A, bone morphogenetic (BMP)-15, regulate zebrafish. Therefore, third major focus test if these factors, as well gonadotropins, mPRs. Overexpression BMP-15 significantly reduced, while knockdown increased, levels. However, remained unchanged overexpression or knockdown. Treatment human chorionic gonadotropin (hCG) resulted an mPRbeta, but not mPRalpha, Activin-A induced a dose- time-dependent manner. On other hand, TGF-beta1 treatment suppressed mPRalpha. Taken together, findings further support role mPRs suggest BMP-15, exert regulatory effects part regulating mPR expression.
