摘要: Fibroblast growth factor 2 (FGF-2) is a trophic expressed by glial cells and different neuronal populations. Addition of FGF-2 to spinal cord dorsal root ganglia (DRG) explants demonstrated that specifically increases motor neuron axonal growth. To further explore the potential capability promote axon regeneration, we produced lentiviral vector (LV) overexpress (LV-FGF2) in injured rat peripheral nerve. Cultured Schwann transduced with added collagen matrix embedding or DRG significantly increased but not sensory neurite outgrowth. LV-FGF2 was as effective direct addition vitro. Direct injection into sciatic nerve resulted expression FGF-2, which localized basal lamina cells. investigate vivo effect overexpression on regeneration after injury, were grafted silicone tube used repair resected Electrophysiological tests conducted for up months injury revealed accelerated more marked reinnervation hindlimb muscles animals treated LV-FGF2, an increase number neurons reached distal tibial at end follow-up.
