Memantine is neuroprotective in a rat model of pressure-induced retinal ischemia.

摘要: PURPOSE. To quantify vitreous amino acid concentrations in pressure-induced retinal ischemia and to evaluate the neuroprotective effect of memantine, a N-methyl-D-aspartate (NMDA) antagonist, administered before at two time intervals after ischemia. METHODS. Retinal was induced 10 rats by elevating intraocular pressure 120 mm Hg. The acids samples were measured high-pressure liquid chromatography. In another series 56 rats, similar fashion. Fifteen received 20 mg/kg day memantine subcutaneous osmotic pump starting 2 days ischemia, 13 intraperitoneally (ip) 0.5 4.5 hours reperfusion, ip 3.5 7.5 15 vehicle alone. Ischemic damage histologically quantified 14 RESULTS. Compared with nonischemic fellow eyes. there an elevation (P 0.05) when injected CONCLUSIONS. increased glutamate glycine. Both agonists NMDA receptor. receptor antagonist reduced ganglion cell loss given systemically or within 30 minutes
