摘要: The time course of BCNU-induced hepatotoxicity was studied in rats for up to 4 months after single p.o. doses. Hepatic function assessed by a series standard techniques. By 1 week all doses BCNU prolonged pentobarbital hypnosis and sulfobromophthalein retention. Later, elevations serum bilirubin appeared, but these were delayed as long 63 days at the lowest dose. Serum direct-reacting early stages later shifted indirect-reacting coincidental with reduction Histopathologic study revealed pericholangitis necrosis bile ductules. biliary hyperplasia cirrhosis developed. These findings disclose unique ability produce progressive bimodal toxicity resulting hyperbilirubinemia.
