摘要: The first report of silencing in the vegetative phase growth fungi was made Neurospora crassa. A loss hygromycin resistance observed as a result transformation with plasmid carrying bacterial phosphotransferase ( hph ) gene, fused to promoter trpC gene Aspergillus nidulans. Growth crassa presence cytosine analog 5-azacytidine that prevents methylation, resulted reactivation suggesting DNA methylation is involved silencing. When both transforming and homologous endogenous sequences —it termed “quelling.” Evidence quelling general mechanism, not limited specific genes Neurospora, provided by fact qa-2 encoding quinic acid dehydrogenase used genetic marker for experiments also silenced. As well being dominant, it demonstrated posttranscriptional mechanism.
