Mass spectrometric measurement of neuropeptide secretion in the crab, Cancer borealis, by in vivo microdialysis

摘要: Neuropeptides (NPs), a unique and highly important class of signaling molecules across the animal kingdom, have been extensively characterized in neuronal tissues various crustaceans. Because many NPs are released into circulating fluid (hemolymph) travel to distant sites order exhibit physiological effects, it is measure secretion these from living animals. In this study, we report on extensive characterization crab Cancer borealis by utilizing vivo microdialysis sample hemolymph. We determined necessary duration for collection samples, enabling more comprehensive identification NP content while maintaining temporal resolution sampling. Analysis microdialysates using hybrid quadrupole-Orbitrap™ Q-Exactive mass spectrometer revealed that than 50 neuropeptides 9 peptide families—including allatostatin, RFamide, orcokinin, tachykinin-related RYamide families – were circulatory system. The presence peptides both as well hemolymph indicates their putative hormonal roles, finding merits further investigation. Preliminary quantitative measurement identified suggested several potential candidates maybe associated with circadian rhythm borealis.
