摘要: Mediator is an evolutionarily conserved multisubunit protein complex that plays a key role in regulating transcription by RNA polymerase II. The functions serving as molecular bridge between DNA-bound transcriptional activators and the basal apparatus. In humans, was first characterized thyroid hormone receptor (TR)-associated (TRAP) facilitates ligand-dependent activation TR. More recently, has been established essential coactivator for broad range of nuclear receptors (NRs) well several other types gene-specific activators. A single subunit complex, MED1/TRAP220, required direct interactions with NRs. coactivates NR-regulated gene expression facilitating recruitment II-associated Importantly, acts concert NR coactivators involved chromatin remodeling to initiate target genes multistep manner. this review, we summarize functional signaling pathways emphasis on underlying mechanisms which interacts NRs subsequently their action.
