摘要: Single actin filaments stabilized with fluorescent phalloidin exhibit ATP-dependent movement on myosin fixed to a surface. At pH 7.4 and 24 degrees C, the rates of average 3-4 micron/s skeletal muscle 1-2 Dictyostelium myosin. These are very similar those measured in our previous assays. The relatively independent type used. filament velocity shows broad optimum between 7.0 9.0, concentration ATP required for half-maximal is 50 microM. Evidence was obtained suggest that over requires at most number heads single thick filament. This system provides practical, quantitative myosin-movement assay purified proteins.
