Effect of inhibition of nitric oxide synthase on chronic tension-type headache: a randomised crossover trial.

摘要: Summary Background Studies in animals have shown that nitric oxide plays an important part central sensitisation and inhibitors of synthase (NOS) decrease models persistent pain. The efficacy NOS has not been tested patients with tension-type chronic headache. We aimed to show whether N G -monomethyl-L-arginine hydrochloride (L-NMMA), inhibitor NOS, is effective relieving pain such patients. Methods undertook a randomised double-blind, crossover trial 16 chronic-tension-type Patients were assigned intravenous infusion 6 mg/kg L-NMMA or placebo on 2 days separated by at least 1 week order. Headache intensity was measured 100 mm visual analogue scale, verbal rating scale baseline 30 min, 60 120 min after start treatment. primary endpoint reduction the active treatment compared placebo. Findings reduced significantly more than placebo: treatment, mean score decreased from 49 33 44 40 (p=0·01). Pain also lower for (p=0·02). Interpretation Inhibition had analgesic effect Further tests are required before clinical application.
