Incomplete Elimination of the ABBOS Epitope of Bovine Serum Albumin Under Simulated Gastrointestinal Conditions of Infants

摘要: OBJECTIVE Because the bovine serum albumin residues 126–144 (ABBOS) have been reported to be responsible for autoimmune reaction directed against pancreatic islet cells, it was our aim study potential survival of ABBOS epitope during digestion in gastrointestinal tract. RESEARCH DESIGN AND METHODS Either nontreated (commercially available) or heat-treated (BSA) hydrolyzed vitro with pepsin at a pH 2.0, 3.0, and 4.0 subsequently enzymes 7.5. Cross-reactivity between peptide BSA hydrolysates determined by competitive enzyme-linked immunosorbent assay (ELISA) using rabbit polyclonal antibody raised BSA. RESULTS Biochemical findings clearly showed that degradation simulated physiological depended on its conformation pepsin-catalyzed hydrolysis. Raising hydrolysis from 2.0 decreased efficiency process, especially when had first denatured heat treatment. As consequence, large proportion cross-reactive anti-ABBOS antibody-binding sites still intact final hydrolysates. CONCLUSIONS The present results suggest will not completely eliminated under conditions prevail stomach infants (pH 3–4).
