Beta 2-adrenoceptor stimulation increases the number of antigen-specific precursor B lymphocytes that differentiate into IgM-secreting cells without affecting burst size.

摘要: Previous studies have shown that early addition of a beta 2-adrenergic agonist to whole splenocyte cultures immunized with SRBC induced an increase in the number cells secreting Ag-specific antibody. Because low frequency B lymphocytes these cultures, it has been difficult determine cellular mechanism by which this is produced. To gain insight into mechanism, present study was designed evaluate responsiveness TNP-specific cultured at both high density and limiting dilution keyhole limpet hemocyanin (KLH)-specific, IL-4-producing Th lymphocytes, TNP-KLH, agonist, terbutaline. The results showed maximal twofold anti-TNP IgM-secreting amount IgM secretion occurred terbutaline-exposed after 5 days bulk culture. This response concentration-dependent manner inhibited concomitant culture beta-adrenoceptor antagonists. No appreciable change measured level either IgG1 terbutaline plus Ag-exposed or MHC class II expression on as compared Ag alone. These data raised possibility 2-adrenoceptor stimulation differentiation larger proportion lymphocyte precursors cells, extensive proliferation constant precursors, both. Limiting differentiated without affecting produced each precursor clone.
