摘要: Abstract Type II alveolar epithelial cells were isolated from fetal rat lung by differential adherence in monolayer culture. The preparation had a high degree of purity, as assessed phase contrast microscopy and immunocytochemistry. Purity, based on reactivity with specific anti-adult serum (SAALS), which recognizes only type cells, was 91% for 19-day lungs 79% 21-day lungs. lower purity cultures derived 1-day postnatal (51% reactive SAALS) is probably due to tendency the neonatal rats adhere culture dishes than rats. contained higher percentage phosphatidylglycerol incorporated [Me-3H]choline faster into phosphati-dylcholine (PC) Moreover, cell preparations at day 21, lamellar bodies 19. observation these differences stage maturation indicates that differences, are typical features original material, not obliterated differentiation during according present procedure capable synthesizing PC disaturated species. This method isolation may be useful tool studies concerning surfactant synthesis its regulation lung.
