La protéine de stress Hsp27 / HspB1, une cible de choix en thérapie anti-cancéreuse

摘要: Hsp27 appartient a la famille des proteines dites de survie comme Bcl2 ou survivine. C’est une proteine anti-apoptotique qui subit deregulation son expression dans nombreux types tumoraux. Elle est caracterisee etant cible therapeutique majeure. Au cours ma these, j’ai isole peptides stabilises, dit aptameres, capables d’inhiber fonctionnellement les activites anti-apoptotiques et tumorigenes d’Hsp27. Ces aptameres perturbent biochimie structurale induisent le blocage du cycle cellulaire in vivo. Parallelement cette etude, caracterise effets depletion sur formation metastases tumeurs osseuses. J’ai aussi montre que modification taux induisait degradation differentes proteines, clientes, caspase3, HDAC6 STAT2.
