摘要: Altered capacity for self-renewal and differentiation is a hallmark of cancer, many tumors are composed cells with developmentally immature phenotype. Among the malignancies where processes that govern cell fate decisions have been studied most extensively acute myeloid leukemia (AML), disease characterized by presence large numbers "blasts" resemble progenitors. Classically, defining properties AML were said to be aberrant block differentiation, term "differentiation therapy" was coined describe drugs promote maturation leukemic blasts. Notionally however, simplistic view such agents "unblock" at odds cancer stem (CSC) hypothesis posits hierarchically organized CSCs, which underpin growth, retain progress more mature state. Herein, we will review recent developments providing unprecedented insights into non-genetic heterogeneity both steady state in response treatment, propose new conceptual framework therapies aim alter cancer.
