摘要: Today, proteomics usually compares clinical samples by use of bottom-up profiling with high resolution mass spectrometry, where all protein products a single gene are considered as an integral whole. At the same time, proteoforms, which considers variety species, offers potential to discover valuable biomarkers. Proteoforms species that arise consequence genetic polymorphisms, alternative splicing, post-translational modifications and other less-explored molecular events. The comprehensive observation proteoforms has been exclusive privilege top-down proteomics. Here, we review possibilities approach address microheterogeneity human proteome. Special focus is given shotgun structure-based bioinformatics source hypothetical can potentially be verified targeted spectrometry determine relevance diseases.
