摘要: Bifidobacteria are claimed to contribute positively human health through a range of beneficial or probiotic activities, including amelioration gastrointestinal and metabolic disorders, therefore this particular group commensals has enjoyed increasing industrial scientific attention in recent years. However, the molecular mechanisms underlying these still largely unknown, mainly due fact that tools for bifidobacteria rather poorly developed, with many strains lacking genetic accessibility. In work, we describe generation transposon insertion mutants two bifidobacterial strains, B. breve UCC2003 NCFB2258. We also report creation first mutant library strain, employing Tn5-based transposome strategy. The was found be composed clones containing single insertions which appear randomly distributed along genome. usefulness perform phenotypic screenings confirmed identification analysis defective D-galactose, D-lactose pullulan utilization abilities.
