摘要: Abstract Objective To establish a high throughput, low cost, and simple nanotechnology-based method for the detection of single nucleotide polymorphism (SNP) loci in type 2 diabetes mellitus (T2DM). Methods Multiplex ligase reaction (LDR) amplification was performed using fluorescently labeled magnetic nanosphere-bound upstream LDR probes downstream with unique fluorescent group each SNP locus. The amplified products were separated by nanospheres then scanned fluorescence spectroscopy. Four associated T2DM detected, including rs13866634 locus SLC30A8, rs10811661in CDKN2A/2B, rs1111875 HHEX gene, rs7903146 TCF7L2 gene. genotype also determined DNA sequencing as control. Results genotypes four gene nanosphere-based multiplex consistent results. accuracy rate 100%. Conclusion A based on PCR established simultaneous susceptibility genes.
