摘要: Gyrase is an essential topoisomerase in bacteria that introduces negative supercoils DNA and relaxes the positive form downstream of proteins tracking on DNA, such as or RNA polymerases. Two gyrase mutants suffer partial loss function were used here to study need for replication restart conditions which activity affected. We show preprimosomal protein PriA viability these gyrB mutants. The helicase not essential. lethality priA double suppressed by a dnaC809 mutation, indicating requirement primosome assembly strains. combination mutations independent level supercoiling, also co-lethal presence DeltatopA mutation. Inactivation homologous recombination did affect mutants, does require formation intermediate. propose replisome disassembled from forks when progression blocked accumulation restarts via PriA-dependent assembly, directly in-activated forks, without
