SEPARATION OF PHOSPHOMOLYBDATE BY AFFINITY CHROMATOGRAPHY

摘要: A new method of separating phosphomolybdate from phosphoric esters and anhydrides using a small column polyvinylpolypyrrolidone is introduced. When mixture phosphate compounds ammonium molybdate (1–3%, pH 3-5) poured onto such column, inorganic orthophosphate (Pi)p1 selectively adsorbed the material as phosphomolybdate, while other compounds, which do not react with molybdate, are drained through column. Using radioisotopes, retention recovery percentages were measured. At 3, 99.99% Pip32 was retained in 97 ± 1% ATPp32 recovered effluent. Retained eluted out later 0.5 M hydroxide percentage 98 1%. Unlike methods separation little affected by presence reducing agents. The use disposable columns, can be prepared easily, packed ahead time, stored for use, makes radioisotopes assay convenient contamination-free.
