IL-13 Overexpression Predisposes to Anaphylaxis Following Antigen Sensitization

摘要: Anaphylaxis represents an extreme form of allergic reaction. This acute-phase component allergy and asthma is triggered by allergen-induced degranulation mast cells following the cross-linking cell surface-bound, allergen-specific IgE, resulting in liberation inflammatory mediators development bronchoconstriction. We used IL-13 transgenic mice to investigate role this Th2 cell-derived cytokine onset disease. Strikingly, IL-13-transgenic were highly predisposed fatal anaphylaxis Ag sensitization. response correlated with substantially elevated levels circulating Ag-specific degranulation, histamine release. Furthermore, allergen exposure also induced phenotypic changes typical asthma, including pulmonary fibrosis, goblet hyperplasia, cytokines, eosinophilia, airways occluded mucus Charcot-Leyden crystals. Expression IL-4 was not required for induction IgE-mediated responses. These data represent first characterization a functional IL-13-induced IgE generation immediate hypersensitivity reactions highlight importance symptoms atopy.
