摘要: A number of requirements have to be met by a plasmid useful cloning vector, suitable for the introduction cloned DNA into bacterial cell. Since efficiency transformation cells drops drastically when plasmids larger than 15 kb are used, size vector should small, preferably 3–4 kb. In this way foreign fragments 10–12 can accommodated. Cloning vectors contain an origin replication that operates in organism which one wishes introduce DNA. Furthermore gene serve as selectable marker been transformed — example, encoding antibiotic resistance. Finally or several unique recognition sites restriction enzymes, not located essential regions plasmid.
