SLE-like autoantibodies and Sjögren's syndrome-like lymphoproliferation in TGF-beta knockout mice.

摘要: Mice bearing the TGF-beta 1 null mutation (-/-) develop lymphoid infiltrates in heart, lungs, salivary glands, and other organs similar to those seen pseudolymphoma of Sjogren's Syndrome. We studied sera from -/- mice found elevated Ab levels dsDNA, ssDNA, Sm ribonucleoprotein. No Abs SSA/Ro or SSB/La no IgM rheumatoid factor were found. Serum autoantibodies predominately IgG specific as shown by ELISA inhibition studies. Antinuclear patterns on Western blots varied one mouse next, indicating a random process responsible for diversity. Wild-type heterozygote had autoantibodies. Ig glomerular deposits mice, that these may be pathogenic. Treatment with dexamethasone failed suppress autoantibody production. These represent an overlap combining autoimmune serology SLE tissue SS. Our results support concept is important naturally occurring immunosuppressive cytokine whose absence can lead systemic disease.
