A Combinational Therapy of Articular Cartilage Defects: Rapid and Effective Regeneration by Using Low-Intensity Focused Ultrasound After Adipose Tissue-Derived Stem Cell Transplantation.

摘要: Although low-intensity pulsed ultrasound has been reported to be potential cartilage regeneration, there still unresolved treatment due fibrosis and degeneration by a lack of rapid high-efficiency treatment. The purpose this study was investigate the effect combination therapy focused acoustic force stem cells at site for fast efficient healing on regeneration. Using rat articular defects model, one million adipose tissue-derived (ASCs) were injected into defect site, (LOFUS) in range 100–600 mV used 20 min/day 2 weeks. All experimental groups sacrificed after 4 weeks total. gross appearance score hematoxylin eosin (H&E), Alcian blue, Safranin O staining measuring chondrogenic potential. characteristics observed, type II collagen, Sox 9, aggrecan, X collagen stained with immunofluorescence. results comprehensive analysis calculated using Mankin scoring method. scores regenerated chondrocyte-like H&E images higher LOFUS-treated compared those negative control or ASC-treated groups. blue demonstrated that 100 300 mV LOFUS showed greater synthesis glycosaminoglycan proteoglycan. ASC + LOFUS group positive regulation 9 aggrecan which indicated occurrence regeneration based result. therapy, involved ASC LOFUS, quickly effectively reduced defects.
