Intracellular trafficking mechanism, from intracellular uptake to extracellular efflux, for phospholipid/cholesterol liposomes

摘要: Liposomes are widely used as drug delivery vehicles to transfer chemotherapeutic agents, proteins, and nucleic acids into target cells. To improve therapeutic effects reduce unexpected toxic side-effects, it is necessary understand the mechanism of liposomal uptake cells, intracellular fate internalized liposomes. The synthesized components in construction liposomes remains unclear. In work presented here, we investigated trafficking processes from extracellular efflux using conventional constructed with phospholipids (DOPC) cholesterols (Chol). Following transport via endocytosis, DOPC was localized endoplasmic reticulum (ER) Golgi apparatus after escape endosome/lysosome, whereas Chol only ER. Moreover, proteins involved were identified. Additionally, showed that partly effluxed ABCG1, while ABCA1 or ABCB1; suggesting each component examined this study through different transporters. Our findings offer valuable information regarding targeted specific organelles, how possibly avoid following multiple applications liposome formulations.
