摘要: Maternal physiological hypercholesterolemia (MPH) allows a proper foetal development; however, maternal supraphysiological (MSPH) associates with endothelial dysfunction and early development of atherosclerosis. MSPH courses reduced endothelium-dependent dilation the human umbilical vein due to nitric oxide synthase activity compared MPH. Whether modifies availability cofactor tetrahydrobiopterin is unknown. We investigated whether MSPH-associated lower vascular reactivity results from bioavailability tetrahydrobiopterin. Total cholesterol <7.2mmol/L was considered as (n=72 women) ≥7.2mmol/L (n=35 women). Umbilical veins rings were used for assays (wire myography), primary cultures cells (HUVECs) measure synthase, GTP cyclohydrolase 1, dihydrofolate reductase expression activity, well content. relaxation caused by calcitonine gene-related peptide, phenomenon partially improved incubation sepiapterin. HUVECs showed (l-citrulline synthesis l-arginine) without changes in its protein abundance, level MPH, reversed Expression 1 MSPH, expression. pathophysiological condition reducing leading NOS endothelium.
