Characterization of immortalized rabbit lacrimal gland epithelial cells.

摘要: To establish an immortalized lacrimal gland epithelial cell line, the orbital glands of normal New Zealand White rabbits were multiply injected with immortalizing amphotropic retroviral vector (LXSN16E6E7) containing E6 and E7 genes human papillomavirus type 16. Lacrimal removed after 2 d acinar cells isolated cultured on Matrigel-coated 60 mm2 plates DMEM-F12 supplemented 5% Nu-serum V. Transformed selected in G418 sulfate for 7 passaged. Morphology was similar to that described both vivo vitro, rough endoplasmic reticulum secretory granules. These characteristics remained unchanged continued exhibit typical polygonal epithelioid structure. The have been maintained culture 14 mo. gone through 58 passages without loss proliferation or characteristics. Immunohistochemistry Western blots showed positive reactivity component, transferrin, transferrin receptor, which are proteins found gland. Functional analysis by stimulation a cholinergic agonist, carbachol (100 µM), resulted significant release protein. This is first report rabbit cell. will provide valuable tool molecular functions.
