Changes in parathyroid hormone receptors during chondrocyte cytodifferentiation.

摘要: The purpose of this study was to investigate the relationship between changes in parathyroid hormone (PTH) receptor levels and chondrocyte maturation during endochondral ossification. Chondrocytes were isolated from growth plate rabbit ribs maintained presence 10% serum mass cultures. Treatment with PTH-(1-84) a PTH-(1-34) fragment suppressed increases alkaline phosphatase activity type X collagen 1 alpha,25-dihydroxyvitamin D3 abolished 45Ca incorporation into mineral, all which occurred parallel untreated cultures hypertrophic (terminal) stage. These effects PTH observed at low concentrations (10(-10) 10(-9) M) within 24-48 h treatment. also increased [35S]sulfate newly synthesized proteoglycans. In contrast, middle carboxyl-terminal fragments tested had little effect on proteoglycan synthesis or terminal differentiation. binding 125I-PTH-(1-34) cells greater than that liver, skin, muscle, brain, kidney. When correlation stage examined, we found its 72-kDa resting proliferating chondrocytes, 10-fold matrix-forming thereafter decreased chondrocytes both vitro situ. Scatchard analysis revealed due number, not affinity, receptor. paralleled those findings suggest stage-dependent PTH/PTH-related peptide localize stimulation inhibition precocious hypertrophy zone plates.
