摘要: Production of biological (protein)-based therapeutics offers fundamental challenges. The ability to generate unique life-saving therapies has been engaged by many commercial concerns, mainly using eukaryotic (mammalian) cell culture platforms. Genes encoding the valuable biopharmaceuticals are introduced into host where they integrate cellular genome. What goes on within nucleus is no longer a black box but it become clear over last decade that we have not yet begun fully appreciate complexity this sub-cellular compartment and there remains significant scope further optimise stage bioprocessing. This review highlights current vision in relation its role as controller expression genes for production desired product. layers interwoven – eu- hetero-chromatin, epigenetic marking genomes, nucleosomes, factories chromosome territories will be described. As knowledge base accumulated led use approaches seek maximise genes, rationalised understanding nuclear architecture higher level genome regulation.
