Overexpression of the laeA gene leads to increased production of cyclopiazonic acid in Aspergillus fumisynnematus

摘要: To explore novel bioactive compounds produced via activation of secondary metabolite (SM) gene clusters, we overexpressed an ortholog laeA, a that encodes global positive regulator metabolism in Aspergillus fumisynnematus F746. Overexpression the laeA under alcA promoter resulted production less pigment, shorter conidial head chains, and fewer conidia. Thin-layer chromatography (TLC) high-performance liquid (HPLC) analysis revealed SM OE::laeA was significantly increased, included new metabolites were not detected wild type. Among them, compound named F1 selected on basis its high levels antibacterial effects. purified by column preparative TLC identified as cyclopiazonic acid (CPA) LC/MS, which had been previously known mycotoxin. As A. to produce CPA, these results suggest overexpression can be used synthesis useful compounds, even fungus for genome sequence is unavailable.
