Chicken GHR antisense transcript regulates its sense transcript in hepatocytes

摘要: An increasing number of evidences indicated that long noncoding RNAs (LncRNAs) regulate a variety biological progresses via different mechanisms. Our previous study had identified chicken growth hormone receptor (GHR) antisense transcript (GHR-AS) which regulated GHR sense (GHR-S) in LMH cells. In the present study, roles GHR-AS and its regulatory mechanism were analyzed hepatocytes. The expression patterns liver GHR-S, Let-7b ascended with development chicken. hepatocytes proliferation was promoted more cells entered into DNA synthesis (S) phase when overexpressed while cell slowed fewer S interfered. Meanwhile, GHR-S increased we it reduced inhibited. S1 Nuclease protection assay formed RNA duplex 3' untranslation regon (3'UTR). or cells, half-time showed delayed trend 5' (5'UTR) overexpressed. Furthermore, level can be decreased by mimics whereas partially rescued co-transfected pGHR-AS 5'UTR mimics. Based on our findings, affected improved stability possibly forming between GHR-AS, competing Let-7b.
