摘要: A biologically based toxicokinetic model was developed to stimulate the metabolic disposition of pyrene in trout with an average body weight 450 g and dosed a single bolus injection chemical (10 mg/kg). The consists membrane-limited muscle compartment six flow-limited compartments including gills, liver, gut, kidney, carcass, blood. are represented by mass balance equations terms for binding tissue blood proteins, biotransformation, penetration rate into muscle, flow rate, mass, etc. also provides nonsaturable saturable clearances liver kidney. Michaelis-Menten constants metabolism (Km, Vmax) were determined from vitro experiments using isolated cells. Renal clearance very close glomerulus filtration trout. Solution system yielded time courses concentration tissues. Predicted concentrations consistent experimental observations at least 6 days. validated comparing predicted results weighing 285 intraarterial dose (3 mg/kg) pyrene. adequate agreement empirical data.
