摘要: Hepatoblastoma (HB) is the most common malignant liver tumor in very young children. Because of multidrug resistance and severe side effects anti-HB drugs during HB chemotherapy, it urgent important to develop new effective therapeutic agents with less toxicity for treatment pediatric patients cancer. Nuclear Receptor Subfamily 5 Group A Member 2 (NR5A2; LRH-1) has been linked various types malignancies, an emerging target many cancers. However role NR5A2 cancer remains elusive. In this study, we found that both mRNA protein expression levels were significantly higher HepG2 HuH6 human cell lines mouse orthotopic than normal tissue. Lentiviral-Mediated Short Hairpin RNA (shRNA) knockdown inhibited proliferation colony formation compared control cells. Furthermore, silencing induced downregulation cycle gene CCND1 regulator c-myc. Similar shRNA, antagonist also (HepG2, HepT1 cells), reduced c-Myc expression. Finally, flow cytometry results showed a decreased percentage cells S phase which resulted significant decrease rate tumorigenicity, increased number G2/M phase, leading arrest at phase.
