摘要: Increasing evidence indicates Chronic Periodontitis (CP) is a comorbidity of Alzheimer's disease (AD), which the most common form age-related dementia, and for latter, effective diagnostic treatment strategies are lacking. Although inflammation present in both diseases, exact mechanisms cross-links between CP AD poorly understood; direct association two has not been reported. This study aimed to identify serum proteins link CP. Two-dimensional differential in-gel electrophoresis was employed analyze samples from 12 patients age-matched controls. Furthermore, determine molecular AD, neuroblastoma SK-N-SH APPwt cells were treated with 1 μg/ml lipopolysaccharide Porphyromonas gingivalis (P.g-LPS). Ten differentially expressed identified patients. Among them, nine up-regulated, one protein down-regulated. Of 10 proteins, five reportedly involved pathology AD: Cofilin-2, Cathepsin B, Clusterin, Triosephosphate isomerase, inter-alpha-trypsin inhibitor heavy chain H4 (ITI-H4). Western blotting indicated significantly higher expression Clusterin lower ITI-H4 group than Control group. The concentration B good correlation MMSE scores. Moreover, level (but that ADAM10 BACE1) increased along prominent increase Aβ1-40 Aβ1-42 cell lysates P.g-LPS-treated cells. inhibition resulted sharp decrease lysates. TNF-α important inflammatory cytokines P.g-LPS-induced upregulation These results show share an association, while could be key diseases. discovery identical provides potential mechanism underlying risk patients, critical elucidating pathophysiology AD.
