摘要: Protoplasts isolated from a barley cell suspension (cv Dissa) were transformed with plasmid DNA containing the neomycinphosphotransferase II (NPT) and β-glucuronidase (GUS) genes, using polyethyleneglycol (PEG) to induce uptake. Transformed microcalli selected in media G418 sulphate. NPT activity was detected all antibiotic-resistant lines, but not NPT-positive lines had GUS activity. Southern analysis confirmed presence of sequences homologous APT genes G418-resistant callus.
