摘要: This work was undertaken to develop a method for the isolation of mitochondria from single cerebral hemisphere in neonatal mice. Mitochondria normal mouse brain isolated by proposed exhibited good respiratory control ratio 6.39 ± 0.53 during glutamate-malate-induced phosphorylating respiration. Electron microscopy showed intact mitochondria. The applicability this tested on naive mice and their littermates subjected hypoxic-ischemic insult. Hypoxic-ischemic insult prior reperfusion resulted significant (p < 0.01) inhibition respiration compared littermates. associated with profound depletion ATP content ischemic hemisphere. expression Mn superoxide dismutase cytochrome C (markers integrity mitochondrial matrix outer membrane) determined Western blot quantity samples. Thus, we have developed adapted may serve as valuable tool study function model immature injury. In addition, suggested enables us examine functional phenotype targeted genetic alteration.
