Long noncoding RNA GAS5 suppresses triple negative breast cancer progression through inhibition of proliferation and invasion by competitively binding miR-196a-5p.

摘要: Triple-negative breast cancer (TNBC) is considered to be the most aggressive and lethal type of cancer. Many studies have suggested that dysfunction long noncoding RNAs (lncRNAs) correlated with metastasis progression. Here, we show levels lncRNA, growth arrest-specific transcript 5 (GAS5), are decreased in TNBC tissues, this down-regulation GAS5 associated an tumor phenotype patients, affecting clinical stage, lymph node overall survival. Using ectopic overexpression system cells, found up-regulation can significantly attenuate proliferation enhance apoptosis cells. Through bioinformatics analysis verification qRT-PCR luciferase assay, bind miR-196a-5p there a negative relationship between expression among patient samples. Furthermore, demonstrated partially undermine promotion effect induced by miR-196a-5p, including invasion downstream FOXO1/PI3K/AKT signal pathway activation. In our study, functioned as competing endogenous RNA (ceRNA) antagonizing miR-196a-5p-expressing These data suggest suppress progression competitively binding therefore may prognostic biomarker TNBC.
