New genetic signals for lung function highlight pathways and pleiotropy, and chronic obstructive pulmonary disease associations across multiple ancestries

摘要: Reduced lung function predicts mortality and is key to the diagnosis of COPD. In a genome-wide association study in 400,102 individuals European ancestry, we define 279 signals, one-half which are new. combination these variants strongly predict COPD deeply-phenotyped patient populations. Furthermore, combined effect showed generalisability across smokers never-smokers, ancestral groups. We highlight biological pathways, known potential drug targets for and, phenome-wide studies, autoimmune-related other pleiotropic effects associated variants. This new genetic evidence has improve future preventive therapeutic strategies
