Efficient shoot organogenesis in petioles of yam ( Dioscorea spp)

摘要: Shoot organogenesis was successfully achieved in petiole explants excised from 6 to 8-week old vitro plantlets of yam Dioscorea rotundata P. cv. Kponan fissa, cayenensis L. Krengle IB14 and IB35, alata Bete bete. Only the basipetal portion acquired competence, plants regenerated within 21 days on MS medium supplemented with 2 % sucrose, 100 mg/l myo-inositol, 10 μM kinetin 1.5 mM putrescine referred as regeneration (YRM). Plant significantly (p < 0.01) higher (42 %) sucrose compared 1, 3 4 which produced 4, 25 15 respectively. The age donor plantlet critical regeneration, highest efficiency obtained plantlets. Addition antioxidants lipoic acid (19.4 μM), l-cysteine (28.5 μM) culture stimulated axillary branching shoots. Among four cultivars tested, bete had similar response at kinetin, while IB35 best a lower concentration (0.5 μM).
