The effect of temperature and shear rate on platelet aggregation.

摘要: Samples of whole blood were obtained from male volunteers and exposed to combinations shear rates temperatures representative cardiopulmonary bypass (CPB) in a modified computer-controlled concentric cylinder rotational viscometer for period 100 s. Blood sampled the chamber was fixed paraformaldehyde, stained with CD41 analysed by flow cytometry. Only platelet-positive particles acquired, each individual cell, or aggregate cells, identified analysis its fluorescence forward light scatter characteristics. Little platelet aggregation observed at less than 4000 s-1 greater 24°C, but large numbers aggregates formed all (p<0.05), more forming 24 30°C 37 42°C (p<0.05). We conclude that process is dependent on both temperature rate. note number platelets become involved under conditions shear-rate typical CPB.
