Betulin suppressed interleukin-1β-induced gene expression, secretion and proteolytic activity of matrix metalloproteinase in cultured articular chondrocytes and production of matrix metalloproteinase in the knee joint of rat.

摘要: We investigated whether betulin affects the gene expression, secretion and proteolytic activity of matrix metalloproteinase-3 (MMP-3) in primary cultured rabbit articular chondrocytes, as well vivo production MMP-3 rat knee joint to evaluate potential chondroprotective effect betulin. Rabbit chondrocytes were reverse transcription-polymerase chain reaction (RT-PCR) was used measure interleukin-1β (IL-1β)-induced expression MMP-3, MMP-1, MMP-13, a disintegrin metalloproteinase with thrombospondin motifs-4 (ADAMTS-4), ADAMTS-5 type II collagen. Effect on IL-1β-induced using western blot analysis casein zymography, respectively. protein also examined . The results follows: (1) inhibited ADAMTS-4, ADAMTS-5, but increased collagen; (2) MMP-3; (3) suppressed These suggest that can regulate secretion, by directly acting chondrocytes.
